摘　　要：免疫刺激复合物(ISCOM)是 Morein 创立的一种疫苗免疫载体,其中的主要佐剂成分是 Quil A(皂素的一种成分).本研究首先用日本血吸虫中国大陆株成虫可溶性租抗原(SWA)作为免疫抗原,按改进方法结合皂素制备 ISCOM,对其在小鼠体内诱导的免疫保护力和 SWA 在 FCA/FIA、蜂胶二种佐剂条件下的免疫保护力进行了比较研究,然后对不同剂量 SWA-ISCOM 的免疫效果进行考察;最后对重组日本血吸虫中国大陆株28kD GST(rSj28GST)-ISCOM 在小鼠体内所诱导的免疫保护力进行观察.结果表明 IS-COM 在进一步应用于各种基因工程苗方面有研究价值。ISCOM is a novel type of immunostimilating complex founded by Morein,which matrix is the glycoside Quil A(a part saponins).In this paper,we mixed soluble worm antigens(SWA)/recombinant 28 kD GST(rSj28GST)of Schistosoma japonicum and saponins to ereat SWA-ISCOM and rSj28GST-ISCOM,and then compared the the adju- vant effects of ISCOM,FCA/FIA,Propolis and PBS(namely on adjuvant)on SWA,at last,we observed the protection of different doses of SWA-ISCOM and the efficacy of rSj28GST-ISCOM.The results were as follows: ①Comparision of the adjuvant effects:Outbreeding Kunming mice were vaccinated twice by I.M.with 50 μg SWA/mouse each time.The worm reduction rates were 18.1%, 13.8%,7.5%,11.4% in groups immunized with SWA-ISCOM,SWA-FCA/FIA,SWA- Propolis,SWA-PBS respectively.The result showed that the efficacy of SWA-ISCOM was better than others,but,the statistical difference was not obvious. ②Comparision of the protection of different doses of SWA-ISCOM:5 groups of outr- eeding Kunming mice were vaccinated three times by I.M.with SWA-PBS and SWA-IS- COM containing 50 μg SWA,100 μg SWA,200 μg SWA,400 μg SWA each time respec- tively.The worm reduction rates were 20.4%,-0.32%,27.8%,23.0%,20.4% respec- tively.In addition,32.2% female worm reduction rate was obtained in mice immunized with SWA-FCA/FIA containing 100 μg SWA,moreover,its statistical difference in female worm burden compared with control group was remarkable. ③Observation of the efficacy of rSj28GST-ISCOM:rSj28GST was a 33 kD fusion pro- tein expressed in Escherichia coli TB1 and purified by glutathion-agoroseaffinity chro- matography,with 3.95 μmol·min^(-1)GST activity detected by 1-chloro-2,4-dinitrobenzene/ glutathion spectrophorotometry.Balb/e mice were vaccinated three times by I.M.with rSj28GST-ISCOM and rSj28GST respectively.The worm reduction rate of rSj28GST-IS- COM group was 25.2%,higher than that of rSj28GST group(15.2%).The antibody lev- els of rSj28GST-ISCOM group were significantly higher than that of rSj28GST group.
