摘  要:用平板计数和聚合酶链反应(PCR)法,检测了暴露于不同温度条件下自然湖水中大肠杆菌基因工程菌(MM3)的存活与衰亡规律。平板计数结果表明,4℃湖水中MM3的平均存活率为35.35%,20℃为22.18%,30℃为20.18%。PCR检测指出,自然湖水温度的高低和暴露MM3周期的长短对检测结果没有影响。用含有氯霉素的LB平板,检测不同温度湖水中MM3的质粒稳定性。おn this study, the survival and death rate of genetically engineered Escherichia coli (MM3) exposed in natural lake water at different temperature was detected by plate counting and polymerase chain reaction. The results of plate counting displayed that, the average survival rate of MM3 in lake water was 3535% at 4℃, and 2218% at 20℃,2018% at 30℃. The result of PCR showed that temperature of natural lake water and exposure time of MM3 didn't influence the detection results.Plasmid stability of MM3 in lake water at different temperature was detected on LB plates containing chloramphenicol, and plasmid loss wasn't found.
