摘  要:目的:对表皮细胞在不同的培养方法、不同的培养液及不同底物中的生长情况进行3方面的探讨,以便选出较好的一种组合进行表皮细胞培养,为改善烧伤皮源不足的问题打下基础。方法:①采用人包皮或体皮制成皮块、皮粒、表皮细胞悬液进行液体培养,比较3种方法中表皮细胞生长情况;②应用纯表皮细胞悬液进行空气液体界面培养,平铺平皿培养及培养瓶液体培养,比较表皮细胞贴壁、生长情况;③另将表皮细胞悬液接种于3种不同的培养液,即RPMI1640,RPMI1640+MEM及MEM比较细胞生长差异。结果:①在适当的条件下,离体的皮肤皮块、皮粒及表皮细胞悬液经培养皆能生长和分化,而表皮细胞悬液生长速度较快,纤维母细胞污染机会最少;②以Biofil为底物的空气液体界面法,细胞生长速度最快,培养出的单层表皮细胞膜片易取出,不收缩;③表皮细胞在MEM,RPMI1640及RPMI1640+MEM3种培养基中的生长情况基本一致。结论:以Biofil为底物的空气液体界面培养法应得到推广应用,它不仅加快了表皮细胞的生长速度,减少了纤维母细胞的污染,易于移植。Objective: To investigate the growing conditions of epidermal cell cultured with different methods, different cultivation liquids and different substrates, so as to choose the best one. Methods:①Skin fragment, microskin and cell suspension of human epithelium were cultured, and the growing conditions were compared. ②Culturing effects of airliquid interface, flatmembrane method and liquid method were compared.③ Culturing effects of different cultivation liquids were compared. Results:①In suitable conditions, three type of epithelium all could grow and differentiate, but the cell suspension growing the fastest and having the least probability of being contaminated with fibroblasts.②The epidermal cell in airliquid interface growing faster than in the other interface, the single layer epithelial sheet cultured from Biofill was easily detached from dish and didn't shrunk. ③The culturing effects of different cultivation liquids were same. Conclusions:The method of using Biofill as substrate withairliquid interface would be widely used for its fast growth rate, free from fibroblast, transplanted simply and its probability of being used in the third degree burn for a long time.
