摘 要:目的:探讨中药生脉注射液对心肌细胞内钙调节功能的影响。方法:采用 Fu-ra-2/AM 荧光标记及同位素^(45)Ca^(2+)负载示踪法测定了心肌细胞内钙浓度及肌浆网摄钙功能。结果:生脉注射液作用早期(10min)能够增加心肌细胞外钙的快相内流,使心肌细胞内游离钙浓度增高,肌浆网摄钙量也有明显增加;生脉注射液作用60min 时,与对照组比较细胞内^(45)Ca^(2+)总量增加,肌浆网对^(45)Ca^(2+)的摄取量明显增高,而细胞浆内游离钙浓度无明显变化。维拉帕米可阻断生脉注射液对细胞内钙的调节作用。结论:生脉注射液通过增加心肌细胞钙内流速度和肌浆网摄钙量,增强了心肌细胞钙贮备能力,进而调节心肌细胞的收缩舒张功能。Objective:To explore the effects of Shengmai injection(SMI)on the myocardial intracellular calcium handling. Methods:The intracellular concentrations of free calcium and the sarcoplasmic reticulum Ca^(2+)(SRCa^(2+)) uptake function were determined using the Fura-2/AM fluorescence and the isotope ^(45)Ca^(2+)loading method.Results:SMI could increase the rapid influx of myocardial extracellular calcium,raise the intracellular concentrations of free calcium,and enhance the SR Ca^(2+) uptake at the early stage (10 min).However,after 60 min of treatment with SMI,the total amount of the in- tracellular ^(45)Ca^(2+) and SR^(45)Ca^(2+) uptake increased whereas the cytosolic concentration of free calcium unaltered significantly compared with the control group.Verapamil hindered the regulatory effect of SMI on the intracellular calcium.Conclu- sion:SMI could enhance the capacity of intracellular calcium reservation by increasing the rate of Ca^(2+)influx and the amount of SR Ca^(2+)uptake and further control systolic and diastolic function of the myocardial cells.
