摘 要:目的:动脉血管平滑肌细胞(VSMC)过度增殖是动脉粥样硬化及冠脉成形术(PTCA)后再狭窄形成的主要原因,为了探讨雷公藤红素预防再狭窄形成的可能,本研究观察了雷公藤红素对培养的大鼠胸主动脉 VSMC 增殖的影响。方法:采用大鼠传代培养的 VSMC(4~7代),将雷公藤红素(0.05~0.40mg/L)加入细胞培养基中,24h 和48h 后用中性红细胞染色法测定细胞总蛋白的方法做细胞计数并测定氚-胸腺嘧啶脱氧核苷(~3H-TdR)掺入的放射性强度每分计数(cpm)值,求各组均值及标准差,采用 q 检验做统计学处理。结果:各药物剂量组的雷公藤均可抑制 VSMC 增殖及对~3H-TdR 的掺入并呈药物剂量依赖性关系,与对照组比较P<0.05或 P<0.01。结论:雷公藤红素能够抑制大鼠 VSMC 的增殖,为该药物可能用于以VSMC 增殖为主的冠心病和再狭窄的形成提供了实验依据。Objective:To explore the possibility of tripterine to prevent restenosis after percutaneous transluminal coronary an- gioplasty.Methods:The 4th to 7th passage culture of aortic smooth muscle cells(SMC)was used,tripterine was added into the culturing medium of SMC in the concentration ranging from 0.05 to 0.40mg/L.The number of SMC was as- sessed by neutral red assay at 24 and 48 hours and the counts per minute (cpm) of^3 H-TdR uptake were determined at 6- hour after the drug was added.Results:Tripterine (0.05-0.40mg/L)inhibited the proliferation and^3 H-TdR uptake of SMC in a dose-dependent manner (P<0.05 or P<0.01).Conclusions:Tripterine inhibits the proliferation of cultured rat SMC which suggests the possibility that tripterine could be a therapeutic agent that inhibits SMC proliferation,and could be a drug for preventing restenosis and atherosclerosis.
