扶正抗癌方对人肝癌细胞分化的影响

摘　　要：目的:寻找新的肿瘤细胞诱导分化剂,探讨扶正抗癌方对人肝癌细胞诱导分化作用及机制。方法:用扶正抗癌方灌胃,分离大鼠血清,作用于体外培养细胞。通过噻唑蓝法、双抗体放射免疫法和酶联免疫吸附法检测,观察扶正抗癌方药物血清对人肝癌细胞SMMC_(7721)分化的影响。结果:扶正抗癌方药物血清处理后,SMMC_(7721)细胞活力明显减弱,细胞内 cAMP 明显升高,细胞分泌白蛋白量增多,分泌甲脂蛋白减少。结论:扶正抗癌方对人肝癌细胞 SMMC_(7721)具有诱导分化作用。其机理可能与升高细胞内 cAMP 有关。Objective:To search for a new differentiation inducer of tumor cells by studying the effect of Fuzheng Kang'ai recipe (FZKAR) on human hepatoma cells and its mechanism.Methods:Serum was taken from the rats which had been treated with FZKAR through gastrogavage.The differentiation-inducing effect of the drug-containing serum on human hepatoma cells (SMMC_(7721)) were observed by means of MTT assay,double antibody radioimmune method and ELISA.Results: After the cells were treated with FZKAR containing serum,the cell vitality was reduced,intracellular cAMP raised,albu- mum-secretion increased,and alpha-feloprotein(AFP) decreased.Conclusion:FZKAR had differentiation-inducing effect on human hepatoma cells,the mechanism might be related to the elevation of intracellular cAMP level.
