摘 要:目的:研究冠心病患者体内的血小板活化状态,建立能准确地反映体内血小板活化水平的实验室方法。方法:测定15例健康志愿者及60例冠心病患者血浆11-去氢-血栓烷B_2岛(11-DH-TXH_2)浓度,同时采用流式细胞术(FCM)和放射免疫法(RIA)测定其血小板GMP-140水平。结果:1.患病组与正常对照组比较,其11-DH-TXB_2浓度明显增高(P<0.01),患病组各组间也有高度显著性差异(P<0.01);用FCM检测GMP-140结果与11-DH-TXB_2变化基本一致;而放免法反映血小板活化状态却不如前两种方法明确。结论:11-DH-TXB_2具有稳定的生物学特性,流式细胞术检测血小板GMP-140特异性强,两者均能准确地反映体内血小板的活化程度。Objective:To study the platelet activity status of patients with coronary heart disease ( CHD) and establish more accurate laboratory methods for evaluating this situation. Methods: Plasma leval of 11 - dehy-drothromboxane B2(11-DH- TXB2) was determined in 15 normal donors and 60 patients with CHD. The a-granule membrane protein ( GMP - 140) of platelet surface was also detected simultaneously with flow cytometry (FCM) and radio - immunity assay (RIA) .respectively. Results:The plasma concentration of 11 - DH - TXB2 in CHD group was significantly increase as compared with those in controls ( P < 0.01). and the 11 - DH -TXB2 leval was obviously different within three subgroups of CHD ( P < 0.01). Though the result of GMP - 140 measured by FCM was consistent with the change of 11 - DH - TXB2, RIA did not assess the platelet activity explicitly. Conclusion: II - DH - TXB2 possesses the character of biological stableness and FCM is specific for determining platelet GMP- 140. So these tests can precisely reflect the degree of platelet activity in vito.
