摘 要:本实验研究了骨髓基质细胞系QXMSC_1体外对CFU-F,CFU-GM,BFU-E及CFU-E集落形成的影响,并通过建立QXMSC_1基质细胞与骨髓造血细胞的长期共培养体系,研究了QXMSC-1在体外促进造血的机理。结果表明,QXMSC_1基质细胞条件培养上清液可促进小鼠CFU-F,CFU-GM,BFU-E及CFU-E集落的生长。在无GM-CSF和Epo存在的情况下,QXMSC_1细胞能维持CFU-GM,BFU-E和CFU-E集落生长。表明QXMSC_1细胞本身可分泌GM-CSF和Epo。QXMSC_1细胞与骨髓造血细胞共培养时,可明显提高CFU-GM集落数。共培养体系中,用微孔滤膜隔断QXMSC_1与造血细胞的直接接触,也能促进CFU-GM集落生成,但比混合培养时低,说明QXMSC_1通过直接接触和分泌细胞因子促进造血的过程。In this report, we studied the effects of bone marrow stromal cell line QXMSCj on the CFU-F, CFU-GM, BFU-E and CFU-E. The mechanism of improving hematopoiesis by stromal cells was investigated by establishing long-term coculture system of QXMSQ stromal cells with the hematopoietic cells of bone marrow. The results indicated that the cultural supernatant of QXMSQ cells could significantly stimulated the growth of CFU-F, CFU-GM, BFU-E and CFU-E. QXMSQ cells could also maintain the growth of CFU-GM, BFU-E and CFU-E with conditioned medium lacking Epo, implying QXMSC1 cells secreted GM-CSF and Epo. In the coculture system, QXMSQ is able to increase the number of CFU-GM. The millipore membrane was used to separate the QXMSQ cells from hematopoietic cells in the coculture, so as to see the way by which the QXMSQ stimulates CFU-GM. The result indicated that the separated QXMSC1 stromal cells are able to support the growth of CFU-GM progenitor, but lower than the contact coculture, implying that the QXMSC1 stromal cell can improve the hematopoiesis by direct contact with hematopoietic cells and secreting cytokines.
