摘  要:血小板生成素(Tpo)是巨核细胞增殖分化和血小板形成的主要调控因子。为了探讨Tpo对CD34^+造血祖细胞的协同扩增作用,我们利用免疫磁珠分离系统(MACS)分选出90.11%-97.57%纯度的脐带血CD34^+细胞,在周的体外液体培养体系中,观察了Tpo与干细胞因子(SCF),IL-3,IL-6,红细胞生成素(Epo),粒-巨噬细胞集落刺激因子(GM-CSF)和粒细胞集落刺激因子(G-CSF)不同组合的扩增效率。结果表明,Tpo具有明显的协同扩增作用,细胞总数、集落形成细胞(CFC)和CD34^+细胞可分别被新增329.50±25.20倍,16.09±3.38倍和11.77±4.78倍,SCF+IL-3+IL-6+Tpo组的扩增作用最强,含Tpo实验组的扩增效率明显高于不含Tpo组;同时,含Tpo的组合还使CFU-MK,CD41a^+细胞,BFU-E和CFU-GM分别扩增了34.67±4.62倍,17.29±2.34倍,14.97±2.89倍和14.46±3.19倍。表明Tpo具有明显的扩增造血细胞的作用和刺激多系造血的活性。Thrombopoietin (Tpo) promises to be a specific and effective stimulator for platelet production. To elucidate the role of recombinant human Tpo in expanding CD34 cells which were purified up to 90. 11 % - 97. 57% by a monoclonal antibody-magnetic microbead sorting system, the effects of Tpo in different combinations of hematopoietic growth factors (HGFs) (including SCF, IL-3, IL-6, Epo, G-CSF and GM-CSF) on the proliferation and differentiation of these cells were studied in a 21-day liquid culture system. A combination of 4 HGFs, including SCF, IL-3, IL-6 and Tpo, was identified as the most potent combination of those tested, and increased total nucleated cells by 329. 50 ± 25. 20 fold, CFC by 16.09 ± 3.38 fold and CD34+ cells by 11.77 ± 4.78 fold, respectively. The combinations of HGFs including Tpo also increased CFU-MK by 34. 67 ± 4. 62 fold, CD41 a+ cells by 11.29 ± 2.34 fold, BFU-E by 14. 97 ± 2. 89 fold and CPU - GM by 14. 46 ± 3. 19 fold, respectively. Our studies indicate that Tpo shows a strong synergic potential for expanding CD34+ cells and hematopoietic progenitor cells of different lineages.
