摘 要:目的:观察内皮素(ET)是否参与糖尿病早期肾脏高灌注、高滤过的形成。方法:应用廓清试验观察4周糖尿病大鼠肾血浆流量(RPF)和肾小球滤过率(GFR);应用半定量逆转录聚合酶链式反应(RT-PCR)分别观察肾皮、髓质ET-1、ET-3、ET-A受体及诱生型一氧化氮合成酶(iNOS)的基因表达情况。结果:4周糖尿病大鼠存在RPF、GFR升高;肾脏皮质ET-1、ET-A受体表达增加,皮质ET-3和髓质ET-1、ET-A受体、ET-3表达无明显变化;肾脏皮、髓质iNOS表达显著升高。结论:内皮素可能不是糖尿病早期肾脏血流动力学异常形成的主要参与因子,皮质ET-1。OBJECTIVE To investigate whether endothelins and Nitric Oxide are involved in the pathogenesis of renal hyperperfusion and hyperfiltration in early diabetic rat kidney.METHODOLOGY Renal plasma flow(RPF)and glomerular filtration rate(GFR)were examined by clearance study.Endothelin(ET)1,ET3,ETA receptor,as well as iNOS mRNA expression were determined by semiquantitative reverse transcription polymerase chain reaction(RTPCR).Ten diabetic rats induced by streptozotocin and twelve control rats were observed for 4 weeks.RESULTS Diabetic rats at the forth week had supernormal RPF(454±024 vs 344±050ml/min·100gwt,P<001)and GFR(115±004 vs 094±003ml/min·100gwt,P<001).Gene expression of ET1 and ETA receptor were found enhanced in the renal cortex while not in the medulla.ET3 mRNA expression is comparable to the control both in the medulla and cortex tissue.iNOS mRNA levels elevated obviously in both diabetic renal cortex and medullaCONCLUSION Renal ET1,ETA receptor and iNOS mRNAs levels were increased in early diabetic rats.
