摘 要:应用油菜素内酯(Brassimolide,以下简称B R)预处理大麦幼苗,对大麦叶向原生质体培养具有良好的生理效应,表现能延长原生质体活力,促进细胞分裂。在相同条件下,BR处理的比对照组的呼吸强度、ATP和可溶性蛋白质的含量均明显提高,从而延缓了大麦叶肉原生质体活力的衰退。试验结果表明,BR预处理大麦幼苗对大麦叶肉原生质体的培养是有益的。In this paper, we studied the effect of BR on the isolation and cell division of barley mesophyll protoplast. In the culture of barley mesophyll protoplast, only pretreatment of BR at the barly seedling period as compared with the control was favourable.we did some research works of physiological activity on the barley mesophyll protoplast. The results showed that the respiratory intensity,ATP content and protein content-all dropped in the general culture and then led to the loss of viability. we found that the BR pretreatment could retard the loss of viability of barley mesophyll protoplast and the respiratory intensity, ATP content and protein of BR pretreatment were much higher than those of unpretreated one. So the BR pretreatment was favourable to the culture of barley mesophyll protopast.
