摘  要:本试验以MS培养基为基本成分,采用不同浓度和组合的2,4-D、KT,NAA和蔗糖进行苜蓿子叶组织培养再生植株过程的研究。筛选获得以MS为基础的、比较快速有效的培养基及其培养方法,即诱导和保持愈伤组织的脱分化培养基(2mg/12,4-D+MS)、诱导分化再生植株的分化培养基(4.0mg/1 kT+20g/1蔗糖+MS)、诱导幼苗生根的培养基(1.0mg/1NAA+10g/1蔗糖+MS),从而使利用MS培养基进行紫花苜蓿组织培养再生植株的研究方法有了一定的改进。在此基础上,进行了22个苜蓿品种子叶出愈率、分化率的筛选试验。结果表明,苜蓿子叶的出愈率受基因型影响较小,而分化率在很大程度上受基因型控制,并从中选出了5个出愈率高、分化力强的品种。Influences of 2,4-D, KT, NAA and sucrose on initation of calli, regeneration of plants and initiation of roots were studied by varying several levels and combinations in media. A effective culture system of Medicago sativa L. has been developed. It consisted of a medium of initiation and maintain for callus (2mg/l 2,4-D+MS, designated as CMS) ; a medium for callus to differentiate into plantlot (4mg/l KT+20g/l sucrose+MS, designated as DMS) ; and a medium for plantlet to induce root (1mg/1NAA+10g/l sucrose+MS, designated as RMS) . Both the initiate capacities of callus and the regenerate capacities of plant (twenty-two alfalfa oultivars)were tested. The results showed, initiato capacity of callus from alfalfa cotyledon was not obviously influenced by genotype, but differentiating capacity for plant regeneration was obviously influenced by genotype. The regenerate frequencies varied with genotyes. Five cultivars with better regenerable genotypes were selected. Key words: Alfalfa, Callus tissue, Genotype, Tissue culture.
