氧化型胆固醇对胰岛细胞活性的损害及诱导凋亡作用

摘　　要：目的探讨氧化型胆固醇(Ch-Ox)中25-羟胆固醇(25-OH)、3β,5α,6β-三羟胆固醇(3-Triol)对体外培养胰岛素细胞活性的损害及诱导凋亡作用,并观察其时效与量效关系。方法体外培养小鼠胰岛素细胞株MIN6细胞,待细胞生长融合后,分别加入不同浓度的Ch-Ox(3-Triol与25-OH)作用24h,或用同一浓度的两种不同Ch-Ox作用不同时间。采用MTT法检测细胞的存活情况,并用TUNEL法检测胰岛细胞的凋亡率。结果Ch-Ox可降低MIN6胰岛细胞活性,并呈时间和剂量依赖性,3-Triol对MIN6胰岛细胞活性影响较25-OH明显;Ch-Ox剂量依赖性影响MIN6胰岛细胞凋亡率,3-Triol诱导凋亡作用较25-OH明显。结论Ch-Ox可剂量依赖性和时间依赖性地降低胰岛素细胞的活性,并剂量依赖性诱导MIN6胰岛细胞凋亡率,提示血液中Ch-Ox浓度升高在胰岛β细胞功能损害中具有一定的作用。Objective To investigate the influence and the apoptosis of MIN6 mouse insulin - secreting cells induced by oxysterols and to observe the time and dosage effect. Methods MIN6 mouse insulin cells were cultured in vitro. After confluence of cells, on one hand, oxysterols （3 - Triol 与 25 - OH） in series concentrations was added to culture medium to act for 24 hours, and on the other hand, oxysterols （3 - Triol 与 25 - OH） in the same concentration was added to culture medium to act for different periods. MTT test was used to evaluate the viability and TUNEL test was used to detect the apoptosis of MIN6 cells. Results While treated with either 3 - Triol or 25 - OH by different dosage（25,50, 100μmmol/L） after 24 hours and at different times（6h,12h,18h,24h） ,the viability of MIN6 cells was significantly decreased in a dosage - dependent and time - dependent manner;Two oxysterols cholesterol, especially 3 - Triol, could induce apoptosis of MIN6 insulin - secreting cells in a dosage - dependent manner. Conclusion Both 3 - Triol and 25 - OH can induce apoptosis and decrease the viability of of MIN6 insulin - secreting cells in a dosage and time dependent manner. It is indicated that elevated oxysterols cholesterol concentration in blood may be one factor involved in the dysfunction of pancreatic β cell.
