摘 要:采用荧光定量PCR方法、酶活性速度测定法,探讨了非酯化脂肪酸(NEFA)对体外培养的牛肝细胞中磷酸烯醇式丙酮酸羧激酶(PEPCK)mRNA丰度及其活性的影响。结果显示,添加1.5 mmol/L和2.0mmol/L NEFA时,PEPCK mRNA丰度显著低于对照组(P<0.05);添加0.5~2.0 mmol/L NEFA时,PEPCK的活性显著(P<0.05)或极显著(P<0.01)降低。结果表明,NEFA对体外培养的牛肝细胞中PEPCK mRNA丰度和PEPCK酶活性均具有抑制作用。To investigate the effects of nonesterified fatty acid(NEFA)on the expression of phosphoenolpyruvate carboxykinase(PEPCK) mRNA and activity of PEPCK in bovine hepatocyte cultivated in vitro,the fluorescent quantitative PCR and the velocity methods were used, respectively. 1. 5 mmol/L or 2.0 mmol/L NEFA reduced significantly the expression of PEPCK mRNA compared with that in the control group without NEFA(P〈0.05),and the activities of PEPCK in 4 treatment groups from 0.5 to 2.0 mmol/L NEFA were significantly lower than that in the control group(P〈0.05 or P〈0.01). The results showed that NEFA in hepatocyte cultivated in vitro could inhibit the expression of PEPCK mRNA and activity of PEPCK.
