具有补肾、健脾功效的中药复方对切卵大鼠肾脏、股骨中VDRmRNA表达的影响

摘　　要：目的:观察补肾方、健脾方对大鼠肾脏、股骨中维生素D受体(vitamin D receptor,VDR)表达的影响,探讨补肾、健脾不同功效的中药复方,是否对机体不同组织器官VDRmRNA的表达存在一定选择性。方法:复制切卵大鼠骨质疏松模型,随机分为假切组、模型组、补肾方组和健脾方组,术后12周开始灌胃,灌胃12周后取材。测定骨密度、血清雌二醇浓度,同时对大鼠肾脏、股骨中VDRmRNA进行半定量。结果:补肾方能提高切卵大鼠骨密度(P<0.05),健脾方能维持大鼠骨密度(P>0.05);补肾方、健脾方均维持了切卵大鼠血清雌二醇浓度水平,上调了肾脏和股骨中VDRmRNA的表达(P<0.01)。结论:补肾和健脾中药都能通过上调切卵大鼠肾脏和股骨中VDRmRNA的表达,而起到治疗骨质疏松症的作用。Objective：To observe the effect of kidney-nourishing and spleen-invigorating Chinese compound formula on the expression of vitamin D receptor（VDR） in kidney and femur of ovariectomized rats so as to investigate whether the prescriptions have different selectivity for VDR mRNA in different organs.Methods：The osteoporosis model was made by ovariectomy operation.The rats were randomly divided into sham-operated group,model group,kidney-nourishing group and spleen-invigorating group.Twelve weeks after the operation,all animals were given intragastric administration for 12 weeks.Then,bone mineral density（BMD） and the level of serum estradiol as well as VDR mRNA in kidney and femur were measured.Results：Kidney-nourishing prescription enhanced BMD（P〈0.05） while spleen invigorating prescription maintained BMD（P〉0.05）.Both the prescriptions maintained the level of serum estradiol and upregulated the expression of VDR mRNA in kidney and femur（P〈0.01）.Conclusion：Kidney-nourishing and spleen-invigorating prescriptions can be used to treat osteoporosis by improving the expression of VDR mRNA in kidney and femur.
