摘  要:利用化学方法合成了一种溴氰菊酯新型半抗原,并通过核磁共振鉴定;用活泼酯法和EDC法将溴氰菊酯半抗原分别与牛血清白蛋白(BSA)和卵清白蛋白(OVA)偶联制备免疫抗原和包被抗原,紫外吸收法鉴定表明吸收峰出现在305nm,发生了红移,计算偶联比分别为7:1和6:1;免疫小鼠后获得抗体的效价为64000,IC50为0.16μg/mL。研究为研制溴氰菊酯农药残留免疫速测试剂,建立免疫速测技术奠定了基础。A novel hapten of deltamethrin was synthesized and its structure was verified by HNMR spectrometry. By using active ester and EDC method, the hapten was conjugated with BSA and OVA to prepare the immune antigen and coating antigen, respectively. Results of the UV absorption spectroscopy showed that the absorption peak was found to have a bathoehromic shift at 305 nm, and the calculated coupling ratio was 7 : 1 and 6 : 1, respectively. After immunizing mouse with the hapten, a high selective antibody against dehamethrin achieved a titer value of 64 000 and an IC5o of 0.16 μg/mL for the target pesticide. The paper discussed the possibility for development of ELISA kit in pesticide residue detection, in order to lay a good foundation for application of the fast-immunization testing method in the future.
