清毒片、养正片对白血病细胞u-PA活性影响的研究

摘　　要：[目的]观察中药清毒片、养正片对白血病细胞u-PA活性的影响。[方法]SD大鼠24只,随机分为4组:空白组用生理盐水,清毒片组用清毒片,养正片组用养正片灌胃,化疗组用阿糖胞苷腹腔注射,观察含药血清对HL-60u-PA活性的影响。[结果]与空白对照组相比较,阿糖胞苷和清毒片、养正片药物血清对HL-60细胞培养液上清中u-PA的活性都有一定程度的影响,而以阿糖胞苷含药血清降低u-PA活性的效果较好。[结论]清毒片、养正片的临床治疗作用可能与其降低患者体内白血病细胞u-PA活性表达水平的作用有关。[Objective]We observed the effect of Qingdupian and Yangzhengpian on the u-PA expression in leukemia cells. [Method] 248 SD rats weigh （210±20）g,half male and half female,divided into 4 groups randomly： blank control（0） group,Qingdupian（Q） group, Yangzhengpian（Y） group and Chemotherapy（H） group. Blank control group were fed physiological saline, Q group were fed Qingdupian juice, Y group fed with Yangzhengpian juice,twice a day, 3.5 days. H group were injected Ara-c through peritoneum, once a day,4 times in all. Draw blood of rats 1 -2 hours after the last feed,extract the serum,and store the serum at 4℃. Treat the cells with RPM1640 containing 10 % every kind of serum for 48 hours, precipitate the ceils; the supernatant fluid was collected respectively for ELISA essay to test the activity of u-PA. [Result]The laboratory research showed that all the serum had inhibitive effect on u-PA activity of the supernatant fluid of HL-60 cells,and Ara-c containing serum had the best effect on declining u-PA activity. [Conclusion]Suggest the clinical therapeutic effect of Qingdupian and Yangzhengpian be related to its declining effect on u- PA activities of leukemia cells.
