摘  要:通过大鼠颅骨极量缺损(Critical size defect,CSD)模型的建立,比较骨膜缝合与否对极量骨缺损修复的差异。手术制造大鼠颅骨极量缺损模型,分为骨膜缝合组(FH)与骨膜不缝合组(BF),分别于2、4、8、12周时进行大体观察、X线检查、病理组织学观察;24 h和2、4、8周进行血清中碱性磷酸酶(ALP)的测定。结果显示,FH组、BF组大体观察没有明显差异;BF组和FH组血清碱性磷酸酶(ALP)均高于正常值,且BF组高于FH组(P<0.01);X光检查,缺损部未见明显的质密度变化及修复作用,BF组和FH组修复效果差异不明显;病理组织学检查表明:BF组和FH组均为膜内成骨,可见极少量的新生骨,并有少量的成骨细胞和胶原纤维。大鼠极量骨缺损模型是评价生物植骨材料修复作用的有效方法,骨膜缝合与否对极量缺损模型的修复意义不大,选择不缝合方式较好。Establish of rat calvarial critical size defect (CSD) model,and compare of periosteal sutured or not to explore whether has difference effect on repairing CSD. Surgical created CSD model,and divided the rat into periosteum suture group (FH) and periosteal not suture group (BF). In vivo bone formation was evaluated by general observation, soft X-ray, pathohistology,alkaline phosphatase (ALP) activity, based on the rat calvarial critical size defect model in 2,4,8 and 12 weeks after surgery. FH, BF generally did not observe significant differences in aspect ; both of BF,FH serum ALP activity were higher than normal,and BF higher than FH (P〈0. 01);X-ray had noobvious density changing and healing in the defect region;pathohistology result indicated:BF,FH were intramembranous ossification,with a very small amount of new bone formation and a smallnumber of osteoblast cells and collagen fibers. Rat calvarial critical size defect model is a fine model to evaluate biomaterial effective on repairing bone defect,and periosteum suture or not have nothing to do with repairing rat CSD,and BF model is proved to be a better chose.
