摘  要:目的:研究间接胆红素在J-Grof法直接胆红素测定中的反应动力学问题。方法:配制胆红素标准液,考察Bu(未结合胆红素或称间接胆红素)在直接反应中其反应率与稀释液酸度、蛋白质基质、预保温时间、偶氮反应时间、反应温度及Bu浓度之间的关系。结果:当稀释液酸度pH在2.0~5.0范围时,间接胆红素在J-Grof法直接反应中反应成直接胆红素的值达一平台。标本在稀释液中预保时间越短,Bu的直接反应的反应率越高,反应温度越高,偶氮反应时间越长,Bu的直接反应的反应率越高。在无稀释液时,在一定温度和偶氮反应时间下,Bu在J-Grof法直接反应中的反应率(x)与其浓度C的关系基本符合x=AC-k。结论:x=AC-k很好地说明间接胆红素在J-Grof法直接反应中的反应率与其浓度的关系。Objective:The kinetic studies of indirect bilirubin in direct reaction by J-Grof method.Method:We studied the effects of the acidity of the diluents,protein matrix,reaction temperature,preincubation time,diazo time and the concentration of Bu on the reactivity of Bu in the direct reaction.Results:The DBIL values of the Bu standard in the direct reaction reaches a plateau in the diluents range from pH2.0 to pH5.0. The reactivity of Bu in direct reaction is higher in short preincubation time than in long preincubation time.The higher the reaction temperature is,the longer the reaction time is,the higher the reactivity of Bu in dircet reaction is.At the same temperature and same reaction time,the relationship of the reactivity( x ) of Bu and the concentration of Bu in the direct reaction without any diluents is as follows: x =A C -k (ie log x =logA-Klog C , r >-0.9928),The constant A is associated with the temperature and the diazo reaction time,and the constant K is associated with the diazo reaction time.Conclusion:The presented empirical equation, x =A C -k predicts the reactivities of Bu in direct reaction ,which were in good agreement with the obtained results.
