摘 要:目的探讨2型糖尿病患者单核细胞TLR4表达的变化及其与血浆炎症因子TNF-α的关系。方法应用实时定量PCR及流式细胞仪检测22例2型糖尿病患者及20例健康对照者外周血单核细胞TLR4 mRNA表达及TLR4阳性单核细胞比率,应用ELISA法检测血浆TNF-α浓度。结果2型糖尿病患者外周血单核细胞TLR4 mRNA相对表达量及TLR4阳性单核细胞比率为3.05±0.56、(14.93±3.48)%明显高于对照组1.48±0.30、(7.79±1.63)%(P〈0.05),血浆TNF-α浓度为(49.29±7.09)ng/L,高于对照组(39.55±5.22)ng/L(P〈0.05),TLR4阳性单核细胞比率与血浆TNF-α浓度正相关(r=0.543,P〈0.05)。结论2型糖尿病患者单核细胞TLR4表达增高,单核细胞TLR4可能参与了2型糖尿病的炎症反应。Objective To investigate changes of toll-like receptor (TLR)4 expression in monocytes from type 2 diabetic patients and the relationship between TLR4 expression and the level of plasma inflammatory cytokine TNF-α. Methods The expression of TLR4 mRNA in monocytes and the percentage of TLR4 positive monocytes were assayed by real-time RT-PCR and flow cytometry. Plasma TNF-α concentration was measured by ELISA. Results The expression of TLR4 mRNA in monocytes,the percentage of TLR4 positive monocytes and plasma TNF-α concentration were higher in type 2 diabetic patients than those in control subjects ( P 〈 0.05 ). In type 2 diabetic patients, the percentage of TLR4 positive monocytes was positively correlated with plasma TNF-α. Conclusion Monocytes TLR4 may participate in the inflammatory response of type 2 diabetes.
