摘　　要：建立了LC-MS/MS法测定人血浆中格列美脲的浓度。采用C18色谱柱,以格列齐特为内标,流动相为2mmol/L乙酸铵溶液(含0.5%甲酸)-甲醇(27∶73),电喷雾离子化源,选择性正离子多反应监测,检测离子分别为m/z491.2→m/z352.0(格列美脲)和m/z324.1→m/z127.2(格列齐特)。结果格列美脲在1～400ng/ml浓度范围内线性关系良好,相对回收率大于99.1%,提取回收率大于95.7%,日内和日间RSD均小于10.1%。A LC-MS/MS method was established for the determination of glimepiride in human plasma. A C18 column was used with gliclazide as the internal standard, using 2 mmol/L ammonium acetate （containing 0.5 % formic acid）-methanol （27 ： 73） as the mobile phase. ESI was applied and operated in positive iron multiple-reaction monitoring mode. The detected ions were m/z 491.2→m/z 352.0 （glimepiride） and m/z 324.1→m/z 127.2 （gliclazide）. The calibration curve was linear in the concentration range of 1 - 400 ng/ml.The relative recovery was over 99.1% and the average extraction recovery was above 95.7 %, The RSDs of inter- and intra-day were less than 10.1%.
