摘 要:目的:观察乌头与半夏间半量、等量、倍量配伍剂量外用对镇痛效应和皮肤局部吸收急性毒性的影响。方法:(1)采用小鼠醋酸扭体法试验观察乌头与半夏配伍剂量的外用镇痛效应;(2)采用家兔皮肤局部吸收急性毒性试验观察乌头与半夏不同配伍剂量外用对皮肤的急性毒性。结果:(1)镇痛效应:乌头∶半夏(1∶0.5)在浓度为66.7%时,对醋酸扭体有明显抑制(P<0.05);乌头∶半夏(1∶1)在浓度为66.7%和33.4%时,均能显著抑制醋酸扭体(P<0.01、P<0.05);乌头∶半夏(1∶2)在浓度为66.7%时亦能明显抑制醋酸扭体(P<0.01)。(2)皮肤的急性毒性:乌头∶半夏大剂量(浓度为78%)动物死亡率:1∶1为3/4、1∶2为3/4、1∶0.5为3/4;乌头∶半夏中剂量(浓度为39%)动物死亡率:1∶1为0/4、1∶2为2/4、1∶0.5为1/4;乌头∶半夏小剂量(浓度为19.5%)动物死亡率:1∶1为0/4、1∶2为1/4、1∶0.5为0/4。结论:镇痛试验结果表明,乌头∶半夏比例以1∶1为好,乌头∶半夏在等量时拟有增强镇痛疗效作用;皮肤急性毒性试验结果表明,乌头∶半夏1∶2毒性较大;乌头∶半夏1∶1毒性较小,乌头∶半夏在等量时拟有相互抑制毒性作用,随着剂量的变化毒性拟有增加的趋势,特别是半夏剂量超过乌头时毒性增加最为明显。Objective: To investigate the analgesia effect and acute dermal toxicity of different combination of Aconite and Pinellia Temata. Methods: ( 1 ) Mouse acetic acid writhing test was used to evaluate the analgesia effect; (2) Rabbit dermal absorption test was used to observe acute dermal toxicity. Results: (1) Analgesia effect: combination dose of Aconite/Pinellia Temata (proportion: 1: 0.5, concentration: 66.7%) obviously inhibited acetic acid - induced mice writhing ( P〈0.05}, which was also seen in proportion 1 : 1 at concentration 66.7% and 33.4% (P〈0.01, P 〈 0.05) as well as proportion 1:2 at concentration 66.7% (P〈0.05) . (2) Acute dermal toxicity after mice were administrated with combination of Aconite and Pinellia Temata in three different proportions (1:1, 1 : 2, 1 : 0.5) : when the dose was high (78 % concentration), the number of deaths observed in three groups ( n = 4) were all 3 ; when the dose medium (39% concentration), the number of deaths observed in the three groups (n=4) was 0, 2, and 0 respectively; when the dose was small (19.5% concentration), the number of deaths observed in the three groups ( n = 4) was 0, 1, and 0 respectively. Conclusion: The analgesic test suggests the best proportion is 1: 1, and there is synergistically analgesic effect while combining Aconite and PineUia Ternata in an equivalent proportion. Acute dermal toxicity test reveals that the proportion of 1 : 2 has a maximal toxicity, whereas the proportion of 1 : 1 shows a minimal toxicity. The equivalent Aconite/PineUia Temata combination shows mutual toxicity inhibition, and the effect is enhanced as the proportion changes, especially when the proportion of Pinellia Ternata is more than that of Aconite.
