摘 要:目的提取分离虎杖药材中的白藜芦醇苷,筛选能转化白藜芦醇苷的菌株。方法虎杖药材通过醇提、液液萃取后,萃取物经大孔吸附树脂和氧化铝柱层析进行纯化;利用二步发酵法筛选白藜芦醇苷的转化菌,用TLC和HPLC法跟踪检测白藜芦醇苷和转化产物白藜芦醇,进而用NMR技术确定转化产物的结构。结果与结论从虎杖药材中提取分离得到含量质量分数为95%的白藜芦醇苷,筛选结果表明丝状真菌总状共头霉3.264(Syncephastrum racem osum3.264)能够高效地将白藜芦醇苷转化为白藜芦醇,并从转化后的发酵液中纯化得到含量质量分数为98%的白藜芦醇。Objective To extract piceid from the rhizome of Polygonum cuspedatum Sieb. et Zucc. and screen the strain which could transform piceid into resveratrol. Methods The rhizome of Polygonum cuspedaturn Sieb. et Zucc. was extracted with alcohol and the filtrate was extracted by ethylacetate. The extract in organic phrase was concentrated and separated on macroporous resin and A12 03 column. Two-step fermentation was utilized to screen the strain which could transform piceid. TLC and HPLC were used to detect the substrate and the product. The structure of the product was identified by its NMR data. Results and conclusions Piceid with 95% (w)purity is obtained from Polygonum cuspedatum Sieb. et Zucc. and Syncephastrum racemosum 3. 264 can transform piceid into resveratrol with high efficiency, which is also obtained from the broth with 98 % (w) purity.
