摘 要:目的:观察Toll样受体8(TLR8)在鸡Ⅱ型胶原诱导的关节炎模型小鼠脾脏单个核细胞中的表达及其与IL-10等细胞因子表达的关系。方法:12只DBA/1J小鼠随机分成造模组和阴性对照组。造模组小鼠采用鸡Ⅱ型胶原诱导,在诱导后的第27天出现关节红肿时处死所有小鼠。用ELISA测定小鼠外周血的TNF-α,HE染色观察小鼠关节的炎症变化,Real-time PCR检测脾脏单个核细胞表达IL-10、IL-17A、IL-1β及TLR8的水平。结果:HE染色结果显示,造模组小鼠关节部位与阴性对照组相比出现明显的炎症细胞浸润;造模组小鼠的外周血血清TNF-α水平较阴性对照组明显升高(P<0.01);模型小鼠脾脏单个核细胞表达IL-10、IL-17A、TLR8和IL-1β的水平也明显高于阴性对照组(P<0.01或P<0.05)。相关性分析表明,TLR8的表达与IL-10的表达呈负相关(r=0.945;P<0.01),而与IL-17A、IL-1β无明显相关性(P>0.05)。结论:TLR8在鸡Ⅱ型胶原诱导的关节炎模型小鼠脾脏单个核细胞中的表达水平明显升高。尽管IL-10的表达水平也高于对照组,但其与TLR8的表达呈现明显负相关。AIM: To explore the expression of Toll-like receptor 8(TLR8)in rheumatoid arthritis induced by chicken Ⅱ collogen in mice and analyze the relation of TLR8 to IL- 10, IL-17A and IL-1β. METHODS: Twelve DBA/1J mice were randomly divided into model group (T) and negative group(NC). The paw swelling was seen at the day of 27 post the first immunization with chicken 11 collogen. The serum TNF-α was detected by ELISA and inflammatory cell infiltration was examined by HE. Real-time PCR was used to analysis the expression of IL-10, IL-17A, IL-1β and TLR8 in spleen mononuclear cells. RESULTS: Severe inflammation was detected in model group mice by histological analysis, which was not seen in negative group. The serum TNF-α in model group was enhanced compared with that in negative group( P 〈 0.01). The expression levels of IL-10, IL-17A, TLR8 and IL-1β in spleen mononuclear cells from model group were increased compared with those from negative group (P 〈 0.01, P 〈 0.05 ). Statistical analysis showed that there was negative correlation between the expression of TLR8 and IL-10( P〈0.01 ), and there was no correlation between the expression of TLR8 and IL-17A or IL-1β( P 〉 0.0.5 ). CONCLUSION: The expression of TLR8 was increased in rheumatoid arthritis-mice induced by chicken Ⅱ collogen, and there was negative correlation between the expression of TLR8 and the expression of IL-10.
