摘 要:目的探讨重庆地区人群血管内皮素-1(endothelin-1,ET-1)基因Lys198Asn、TaqⅠ酶切位点基因多态性与原发性高血压(essential hypertension,EH)的关系。方法运用序列特异引物聚合酶链反应(sequence specific primer poly-merase chain reaction,SSP-PCR)和限制性内切酶片段长度多态性(restriction fragment length polymorphism,RFLP)方法分析86例EH患者和138例非EH患者的Lys198Asn和TaqⅠ2个位点基因多态性。结果体质量指数(body mass index,BMI)<25 kg/m2人群中高血压组的TaqⅠ酶切位点基因型分布频率为:GG型26例(72.2%),GA+AA型10例(27.8%);对照组:GG型28例(50.0%),GA+AA型28例(50.0%)。Logistic回归分析显示BMI<25 kg/m2人群中携带GG基因型(TaqⅠ酶切位点)的患者发生高血压的风险增加(P=0.037,OR=2.597)。线性回归结果显示收缩压(SBP)在BMI<25 kg/m2人群的GG型平均值为145.06,显著高于GA+AA型平均值131.97(P=0.018)。结论在BMI<25 kg/m2人群中TaqⅠ酶切位点GG基因型更易导致SBP升高,GG基因型可能是BMI较小人群血压升高的一个危险因素。Objective To investigate the relationship between Lys198Asn and Taq I polymorphisms of endothelin-1 gene and essential hypertension in Chongqing residents. Methods Polymorphisms of Lys198Asn and Taq I were detected in the blood samples fiom 86 patients with essential hypertension (HT) and 138 nor- motensive controls (NT) by sequence specific primer-polymerase chain reaction (SSP-PCR) and PCR-restfie- tion fragment length polymorphism (PCfI-RFLP) Results In the BMI 〈25 kD/m2 subjects, the frequency of GG and CA + AA of Taq I was 26 (72.2% and 10 (27. 8% ) respectively in hypertensive group, 28 (50. 0% ) and 28 (50.0%) respectively in normotensive group. Logistic regression model analysis showed that the GG carriers of Taq I had a significant high risk to develop hypertension (P = 0. 037, OR = 2. 597) in BMI 〈 25 kg/m2 subjects. The liner regression model analysis demonstrated that there was a significant difference in systolic blood pressure (SBP) between CG and CA + AA in BMI 〈 25 kg/m2 subjects (P = O. 018). SBP of the GG carriers (145.06 mmHg) were 13.09 mmHg higher than those of CA + AA carriers (131.97 mmHg). Conclusion Among the BMI 〈25 kg/m2 subjects, those with GG carriers of Taq I might be prone to have higher SBP. The GG genotype might be a risk factor for hypertension in normoweight subjects.
