摘  要:目的比较黄芪、水蛭及其配方含药血清对大鼠系膜细胞生长周期、凋亡的影响。方法制备水蛭高浓度(水蛭组)、黄芪高浓度(黄芪组)和水蛭高黄芪低浓度(配方组)含药血清,并对大鼠系膜细胞(MC)进行干预培养,应用流式细胞仪检测MC生长周期与凋亡,显微镜观察凋亡细胞的形态。结果各组含药血清均能使大量MC处于G0/G1期,与对照组比较差异有统计学意义(P<0.01);各组亦均能提高大鼠MC凋亡率,水蛭组、配方组与对照组比较差异有统计学意义(P<0.01)。结论水蛭、黄芪及其配方含药血清均能阻止MC进入S期从而达到抑制增生的目的,且能提高MC的凋亡率。Objective To study the effect of the serum contained Radix Astragali, Hirudo or their compound on the growth cycle and apoptosis of rat mesangial cells (MC). Methods The serum contained the extract of high-concentration of Hirudo, the serum contained the extract of high-concentration of Radix Astragali, and the serum contained the extract of high concentration of hirudo and the extract of tow-concentration of Radix Astragali were prepared. The cultured MC were measured by flow eytometer to observe the changes in their growth cycle and apoptosis. The morphological changes of apoptotic cells were observed. Results The medicinal serums can keep tbe most MC in the stage of G0/G1, and the significant difference was existing in comparison with that of the control group (P〈0. 01 ). The apoptosis rate of MC in the medicinal groups was increased, and the difference was shown in the Hirudo and compound groups in comparison with that of the control group (P〈C0.01). Conclusion The medicinal serums contained Hirudo and Rudix Astragali and their compound respectively can prevent MC entering the stage of S to inhibit their growth, and they can increase the rate of apoptosis of MC as well.
