四君子汤对脾虚模型大鼠左氧氟沙星药动学影响的实验研究  被引量:4

摘  要:目的:研究四君子汤(人参、白术、茯苓、甘草)对脾虚模型大鼠左氧氟沙星药动学的影响。方法:24只大鼠随机分为4组,生理盐水组(给予生理盐水)、生理盐水+四君子汤组(给予生理盐水和四君子汤)、利血平组(给予利血平预处理)、利血平+四君子汤组(给予利血平预处理后用四君子汤治疗),以20 mg/kg灌服左氧氟沙星,于给药后不同时间点采集血样,用高效液相色谱法分析左氧氟沙星的血药浓度,根据血药浓度-时间数据计算药动学参数。结果:利血平引起的大鼠症状类似于中医临床脾虚证。左氧氟沙星在生理盐水组、生理盐水+四君子汤组、利血平组、利血平+四君子汤组大鼠体内的药动学参数分别为:AUC(0-∞)=(8.55±0.99),(7.41±1.39),(4.68±0.95)和(7.89±1.41)mg/(L.h),Cm ax=(3.31±0.63),(2.38±1.15),(1.29±0.45)和(3.35±1.15)mg/L。与生理盐水组相比,利血平组大鼠体内左氧氟沙星的AUC(0-∞)和Cmax显著降低(P<0.01),与利血平组相比,利血平+四君子汤组大鼠体内左氧氟沙星的AUC(0-∞)和Cmax显著增加(P<0.01),而生理盐水组与生理盐水+四君子汤组参数之间没有显著性差异。结论:脾虚状态下左氧氟沙星的生物利用度显著降低,四君子汤可改善脾虚状态下左氧氟沙星的吸收。AIM: To investigate the influence of Sijunzi Decoction( Radix et Rhizoma ginseng, Rhizoma atractylodis macrocephalae, poria, Radix et Rhizoma glyeyrrhizae) on the pharmacokinetics of Levofloxacin (LVFX) in rats with deficiency of spleen. METHODS : Twenty-four rats were randomly divided into four groups : NS ( normal tion). After a single oral administration of LVFX 20 mg/kg, blood samples were collected at different intervals. The concentrations of LVFX plasma were determined by HPLC. Pharmacokinetic parameters were determined from the plasma concentration-time data. RESULTS: Reserpine led to the syndromes similar to the deficiency of spleen, a traditional Chinese medicine syndrome. The pharmaeokinetic parameters of LVFX in NS, NS + SJZT, R and R + SJZT groups were as follows: AUC(0→∞) = (8.55 ±0.99), (7.41 ±1.39), (4.68 ±0.95) and (7.89 ±1.41)mg/(L · h), respectively and Cmax = (3.31 ±0.63), (2.38 ±1.15), (1.29 ±0.45) and (3.35 ±1.15) mg/L, respectively. Compared with the parameters of LVFX in NS group, Reserpine markedly decreased AUC(0→∞) and Cmax of LVFX(P 〈0.01 ). Compared with the parameters of LVFX in R group, Sijunzi Decoction markedly increased AUC(0→∞) and C max of LVFX (P 〈 0.01 ). However, there was no significant difference in the parameters between NS group and NS + SJZT group. CONCLUSION: The oral hioavailability of LVFX decreases significantly in spleen deficiency rats, whereas Siiunzi Decoction could improve the absorption of LVFX.
