摘　　要：目的:研究冠心病不同中医证型的体内物质基础以探索中医分型的科学内涵。方法:采用基于GC-TOF/MS测定技术的代谢组学研究手段对痰阻心脉和气阴两虚型冠心病患者血浆中小分子化合物进行全面检测,利用多变量数据分析和方差分析等统计方法研究血浆内源性化合物在各组间差异。结果:代谢组学分析不但可把痰阻心脉型和气阴两虚型冠心病患者区别分开,还可以与健康组明显区分。利用PLS_DA建立的痰阻心脉和气阴两虚组数学模型参数与临床中医症状分级量化指标具有显著的相关性。两证型间多个内源性代谢物的含量存在明显差异并与能量代谢、氨基酸代谢和核苷酸代谢异常相关。结论:代谢组学的整体数据和标志物信息均显示出痰阻心脉和气阴两虚型冠心病的差异,提示代谢组学所研究的体内小分子化合物可能正是中医分型的物质基础。To investigate on the pharmacological materia of Traditional Chinese Medicine （TCM） patterns of coronary artery disease for revealing the essence of ＂Bian Zheng （pattern identification）＂, in the present study GC/TOF-MS based metabonomic method was utilized to profile plasma endogenous metabolites of patients with ＂Tan Zu Xin Mai Zheng （phlegm obstructing heart and vessels pattern）＂ and ＂Qi Yin Liang Xu Zheng （qi and yi vacuity pattern）＂. Significant metabonomic difference was observed between the two TCM pattern groups, patients and the healthy control, and the three groups were distinguished clearly by pattern recognition. Very good correlativity was observed between clinic TCM pattern index values and parameters in PLSDA model of the two TCM pattern groups. The endogenous compounds differentiating the two TCM patterns were involved in perturbation in energy, amino acid and nucleotide metabolism. In general, the whole model and biomarkers differentiated and characterized the two TCM patterns, suggesting that metabonomic phenotype, i.e., the endogenous compounds, be the pharmacological materia of TCM patterns.
