摘　　要：目的:建立HPLC-MS测定大鼠血浆中去甲斑蝥素(NCTD)浓度的方法,在此基础上对NCTD在大鼠体内的药代动力学进行初步研究。方法:采用HPLC-MS法,使用DiamonsilTMC18色谱柱和电喷雾电离负离子源,流动相为甲醇-水(20∶80,含2.5%甲酸),流速为0.5ml/min。结果:NCTD在0.53～21.20μg/ml范围内线性关系良好(R2=0.9995),血浆检测限为50ng/ml;日内、日间RSD均<5%。结论:本法快速、准确,适用于NCTD药代动力学的研究;NCTD在大鼠体内的药代动力学过程符合二室模型。Objective：HPLC-MS method was presented for the determination of norcantharidin in rat plasma.Methods：The analysis involved a column of Diamonsil^TM C18 with the negative electrospray ionization.The mobile phase was con-sisted of methanol and water（20∶80, 25% methanoic acid included）.Results：The linear range was 0.53-21.20 μg/ml（R^2= 0.999 5）, intra-day and inter-day precisions over this range RSD〈5.0%.Conclusion：The method is rapid and accurate, and suit the pharmacokinetic study of norcantharidin.Pharmacokinetics of norcantharidin in rats fit the 2-compartment open model.
