摘  要:GA20-氧化酶(GA20-ox)是赤霉素生物合成中的限速酶,它能够催化从GA12到GA9及GA53到GA20过程中的3步氧化反应。笔者以板栗(Castanea mollissimaBlume)为材料,应用基因组步移技术克隆GA20-ox基因,获得长度为2 315 bp DNA序列,其中含有一个GA20-ox基因的全长编码区序列,命名为CmGA20ox1。Cm-GA20ox1长度为1 140 bp,且编码区内存在2个内含子,其推定蛋白CmGA20ox1序列长度为379个氨基酸,氨基酸序列比对分析显示CmGA20ox1与山毛榉坚果的FsGA20ox1(AJ420192)相似性为91.03%,系统发育树分析显示CmGA20ox1与山毛榉坚果FsGA20ox1(AJ420192)遗传距离最近。Gibberellin 20-oxidase(GA20-ox)is an enzyme that can catalyses the three steps ,i. e. oxidization from GA12 to GA9 and GA53 to GA20 in the synthesis of active GAs and is a key point in the regulation of GA biosynthesis. Genomic walking was used to isolate GA20-ox gene from chestnut (Castanea mollissima Blume)and DNA sequence of 2315 bp was obtained, including entire coding region of one GA20-ox gene that was designated as CmGA20ox1. The entire coding region of CmGA20ox1 was 1 140 bp, including two introns or a deduced protein CmGA20ox1 of 379 amino acids,which had 91.03% similarity to FsGA20ox1 (AJ420192) from European beechnut (Fagus sylvatica L. ) analyzed by the amino acid sequence alignment, which showed the nearest genetic distance with FsGA20ox1(AJ420192)by the phylogenetic tree analysis.
