溴氰菊酯残留检测ELISA试剂盒的研制

摘　　要：在前期建立溴氰菊酯间接竞争ELISA检测方法的基础上，研制组装出溴氰菊酯残留检测ELISA试剂盒，并以苹果为试样，对该试剂盒的灵敏度、精密度、准确度、特异性进行参数测定。本试剂盒的IC50为3．999-7．709μg／mL，检出限为0．011-0．024mg／kg，检测范围为0．015～100μg／mL，重现性较好，特异性强，以0．1，0．5，1．0mg／kg3个水平添加苹果，回收率为86．2％～105．8％，变异系数为6．0％～9．8％，接近气相色谱法的检测水平，可满足溴氰菊酯残留初筛检测需要。该试剂盒的成功研制为实际样品检测提供理论基础。On the base of IC-ELISA established, the sensitivity, precision, accuracy and specific of the kit were tested on apple samples. The technique parameters were as follows. IC50 ranged from 3. 999μg/mL to 7. 709 μg/mL; the limits of detection ranged from 0. 011 mg/kg to 0. 024 mg/kg,the detection range was from 0. 015μg/mL to 100 μg/mL. When the spiked concentrations of deltamethrin were 0.1 mg/kg , 0.5 mg/kg and 1.0 mg/kg in apple, the recovery of deltamethrin was 86.2%-105.8% with coefficients of variation of 6.0%-9.8%. The results were close to gas chromatographic（GC） detection.
