柔肝抑纤饮抗DMN肝纤维化大鼠肝窦毛细血管化的实验研究

摘　　要：目的:以复方鳖甲软肝片为对照,观察柔肝抑纤饮抗大鼠肝窦毛细血管化的作用并比较两种药物的疗效差别。方法:建立DMN大鼠肝纤维化模型,将动物随机分组:A组(正常组);B组(模型组);C组(柔肝抑纤饮组);D组(复方鳖甲软肝片组)。分别灌服相应药物。检测血清HA、col-Ⅳ,行胶原染色(Masson)染色、免疫组化染色观察并测定肝组织vWF、LN的表达,电镜观察窦周隙超微结构。结果:C组在降低血清HA、col-Ⅳ水平,减少胶原染色百分比含量以及LN、vWF免疫组化阳性面积等方面,作用明显优于D组(P<0.05);电镜观察发现,C组肝窦内皮失窗孔和内皮下连续性基底膜的形成等病变也相对为轻。结论:柔肝抑纤饮能显著改善LN、vWF等反映肝窦毛细血管化的敏感指标,一定程度上逆转DMN大鼠肝窦毛细血管化,整体疗效优于复方鳖甲软肝片。Objective：To observe the effects of Rougan Yixianyin decoction in preventing the form of sinusoid capillarization caused by injection of DMN, and compared the effects with Fufang Biejia Ruangan pill which was took as the control - drug. Methods ： 48 rats were divided into 4 groups after peritoneal injection of DMN ： group A （ Normal control group） ,group B（ model group）,group C （Rougan Yixianyin group）and group D （Fufangbiejiaruangan pill group）, at the end of the 4th weeks, group C were fed with Rouganyixianyin decoction everyday（ 13.6mL/kg）, group D with Fufang Biejia Ruangan pill（10.88mL/kg）, group A,B with physiological saline respectively for 14 days. Rats of each group were put into death on the 43th days,tested the contents of HA and col - Ⅳ in serum. The histopathology of hepatic tissue was observed too （ by Masson dye, electron microscope, and immunohistochemical methods ） ; the expression of LN and vWF were measured by quantitative analysis. Results：In terms of lower HA and col - Ⅳ in serum, and the masculine area of LN and vWF, there were significant difference in group C than group D（ P 〈 0.05） ;the damage of sinusoid -lining endothelium cells and basal lamina in group C were no more obviously than that in group D too. Conclusion：Rougan Yixianyin decoction has confident effects on preventing the form of liver fibrosis and the form of sinusoid capiUarization caused by DMN, and its curative effects were much better than Fufang Biejia Ruangan pill.
