紫杉醇与白藜芦醇对人胃癌MGC803细胞蛋白表达及其细胞周期的作用

摘　　要：目的:实验研究紫杉醇(PA)、白藜芦醇(Res)合用对人胃癌MGC803细胞影响。方法:通过流式细胞仪(FCM)检测细胞周期,Western blot。方法:研究PA与Res合用前后对体外培养的人胃癌细胞MGC803对细胞的影响。结果:FCM显示细胞生长停止在S期,S期数分数为0.52,抑制细胞有丝分裂及DNA合成。Res与PA合用可以下调Bcl-2表达,上调Bax表达,对P-gp作用不明显。结论:PA与小剂量化疗药物Res合用,可获得化疗药物单独用药相同疗效下调Bcl-2表达,上调Bax表达,增加敏感性,减少药物副作用。Objective： The effect on MGC803 cells from the resveratrol combined with paclitaxel （PA） was studied. Methods ： Flow eytometry was applied to study the effectsof the drugs on the cells cycle of the cell lines. Western blot was used to investigate the eytotoxieity of the drugs against human gastric carcinoma cell line in vitro MGC803 individually or in combinations. Results ： The growth of the cells stopped at period S. The mitosis and the formation of DNA were res trained. PA significantly decreased the survival level of the cells with Res compared with that used individually, down regulation of Bel- 2 ,up-regulation of Bax ,contribution in nubibus of P- gp. Corwlusion：PA could increase the sensitivity of carcinoma cells to kinds of Res,down regulation of Bc1-2 ,up-regnlation of Bax,contribution in nubibus of P- gpand so reduce their side effects.
