涤痰开窍解郁汤对脑卒中后抑郁症的实验研究

摘　　要：目的:探讨中药制剂涤痰开窍解郁汤对脑卒中后抑郁(post-stroke depression,PSD)的大鼠模型行为学能力及脑内神经递质的影响。方法:采用双侧颈总动脉永久性结扎后予以行为限制制作PSD大鼠模型,随机分为模型组、涤痰开窍解郁组、氟西汀组,并设假手术组,采用RP-HPLC-荧光检测法测定大鼠海马区单胺类神经递质的变化,并观察大鼠自发性行为改变。结果:模型组大鼠行为能力下降(P<0.01or0.05),海马区5-HT、5-HIAA、DA、NE水平下调(P<0.05)。盐酸氟西汀、涤痰开窍解郁汤治疗后大鼠的水平活动、垂直活动及清洁活动均较模型组有改善,脑内单胺类神经递质水平提高(P<0.05)。结论:大鼠双侧颈总动脉永久性结扎结合行为限制可为卒中后抑郁的实验及临床研究提供较为理想的动物模型。涤痰开窍解郁汤可以改善卒中后抑郁大鼠的行为及脑内单胺类神经递质水平。Objective： To object the effect of the ditankaiqiao-tang on monoamine neurotransmitter and behavior in the post - stroke depression rats. Methods ： A PSD rat model was set up with permanent bilateral common carotid arteries ligation and a 7 -day＇s limitation. Animal behavior was observed, the neurotransmitter content in the hippocampi area was detected by Reversed-phase HPLC-fluorescence analysis. Results：Both the ability of behavior and the level of NE, DA ,5 - HT,5 -HIAA of the model group decreased in comparison to the sham operated group（ P 〈 0. 05 ）. Compared with model group, the ability of behavior was improved and the level of 5 - HT,5 - HIAA increased in both ditankaiqiao-tang group and fluoxertine hydrochloride group （ P 〈 0. 05 ）. There were no obviously difference between the two groups （ P 〉 0. 05 ）. Conclusion：The ditankalqiao-tang could regulate the neurological function.
