活血潜阳方对高血压大鼠血清AngⅡ MMP-3 TIMP-1含量的影响

摘　　要：目的:观察活血潜阳方对自发性高血压大鼠(SHR)血清AngⅡ、MMP-3、TIMP-1含量的影响。方法:72只雄性10周龄SHR大鼠随机分为4组:SHR对照组、活血潜阳方组、缬沙坦组、活血潜阳方与缬沙坦合用组。另以20只同龄雄性京都W istar(WKY)大鼠作为正常对照组。分别于7周和14周后用酶联免疫法测定血清AngⅡ、MMP-3、TIMP-1含量。结果:SHR血清AngⅡ含量、MMP-3和TIMP-1含量均高于同龄WKY大鼠(P<0.05或P<0.01);活血潜阳方治疗7W有降低SHR血清AngⅡ含量、MMP-3和TIMP-1含量的趋势(均P>0.05),治疗14W可明显降低SHR血清AngⅡ、MMP-3和TIMP-1含量(P<0.05或P<0.01),可使SHR血清MMP-3和TIMP-1含量接近WKY水平(P>0.05)。缬沙坦治疗14周可升高SHR的血清AngⅡ含量,降低血清MMP-3和TIMP-1含量,较疗程7周更为明显。活血潜阳方与缬沙坦合用在治疗14周后明显降低SHR血清MMP-3含量(P<0.05),接近WKY水平(P>0.05)。结论:长疗程(14W)的活血潜阳方治疗可降低SHR血清AngⅡ、MMP-3、TIMP-1含量,从而调控心肌间质重构,是其改善高血压左室肥厚的可能机制。Objective：To explore the effects of Huoxue Qianyang formula on content of angiotensinⅡ,MMP-3 and TIMP-1 in blood serum of Spontaneous Hypertension Rat（SHR）.Methods：Seventy-two SHRs were randomly devided into SHR control（SHR-C） group,Huoxue Qianyang formula（SHR-H） group,Valsartan（SHR-V） group and combination of Huoxue Qianyang formula and Valsartan（SHR-HV） group.Twenty Wistar-Kyoto（WKY） rats served for normal controls.The half of rats in every group were observed respectively for seven or fourteen weeks.After different courses of treatment,content of angiotensinⅡ,MMP-3 and TIMP-1 in blood serum were measured.Results：Serum content of angiotensinⅡ,MMP-3 and TIMP-1 were higher in the SHR-C group than those in the WKY group.They were lower in SHR-H group than those in the SHR-C group after fourteen weeks.Serum content of MMP-3 and TIMP-1 were lower in SHR-V group too,but angiotensinⅡ was higher in SHR-V group than that in SHR-C group,and the effects were more evidently after fourteen weeks than seven weeks.Serum content of MMP-3 in SHR-HV was the lowest in all SHR,and was close to the content in WKY.Conclusion：Huoxue Qianyang formula can lower serum content of angiotensinⅡ,MMP-3 and TIMP-1 of SHR,and ameliorate reconstitution of interstitial substance of LV.It can be one of mechanism which Huoxue Qianyang formula reverse left ventricular hypertrophy in essential hypertension.
