抗人精子外套抗原单抗的纯化及相应抗原的提纯和鉴定

摘　　要：目的：研究人类精浆中一类特异性的抗原成份（精子外套抗原sperm－coatingantigens，SCA）。方法：本实验用已制备得的一株抗人精浆来源的精子外套抗原单克隆杂交瘤细胞株D1．在小鼠腹腔增殖生产大量腹水单克隆抗体，通过快速蛋白液相层析仪（FPLC）系统的羟基磷灰石柱，一次性从小鼠腹水中纯化此抗体，并制备免疫亲和层析柱，从精浆中分离到相应的抗原成份。结果：经鉴定该抗原成份分子量为20kD，来源于精囊腺，具有器官特异性。结论：该SCA的提纯，为抗精子避孕疫苗的研制提供了基础实验材料，并且有助于进一步研究其在生殖活动和免疫不孕病因中的作用。Objective To investigate the sperm-coating antigens(SCA ) in human seminal plasma(HSP). Methods: Monoclonal antibody(McAb IgM) to HSP-specific antigen was prepared by using a hybrid cell line D1 andpurified with hydroxyapatite(HA) on Fast Protein Liquid Chromatography(FPLC) system with one step and aMcAb-bound Sepharose 4B column was prepared. The related HSP-antigen to M.Ab was obtained by immuneaffinity chromatography. Results: The purified antigen mainly gave a single band on SDS-PAGE in a positioncorresponding to a molecular weight of 20 kD, and was proved to be a HSP-specific antigen. Conclusion: Thispurified SCA may be used for preparation of anti-sperm immunocontraceptive vaccines, and it can also be used tostudy reproduction and immunological sterility.
