摘  要:用酵母系统筛选微生物产生的免疫抑制活性化合物,从300多株放线菌中筛选出阳性菌株SIPI-97-84,对其代谢产物进行初步研究,用丙酮浸泡、溶媒萃取、硅胶柱层析和重结晶等方法,从其菌丝体中分离出活性成分SIPI-84-15。通过元素分析和质谱,确定其分子式为C28H31NO6(分子量:477.2145);综合紫外光谱、红外光谱、1H-NMR、13C-NMR等图谱数据,确定SIPI-84-15的结构为含有硝基的多烯化合物,该结构与文献报道的spectinabilin一致,化学命名为:2-甲氧基-3,5-二甲基-6-[四氢-4-[2,4,6-三甲基-7-(4-硝基苯基)-2,4,6-庚三烯亚基]-2-呋喃基],4H-吡喃-4-酮。Using a method for screening immunosuppressive compounds from microorganisms in a yeast system,a positive strain named SIPI9784 whose metabolites exhibited inhibitory activities on yeasts was obtained from more than 300 Actinomycetes collected in our laboratory.Physiologically active compound SIPI8415 was isolated from the mycelia of Streptomyces sp. 9784 by acetone soaking, solvent extraction, silical gel chromatography and recrystallization etc.. By elemental analysis and MS (FAB), its formula was determined to be C28H31NO6(MW: 477.2145); based on the UV, IR, 1 HNMR, 13 CNMR, HMQC, HMBC etc. spectral data and physicolchemical properties of SIPI8415, the structure of SIPI8415 was elucidated and was identical with that of spectinabilin, its chemical name is 2methoxy3,5dimethyl6[tetrahydro4[2,4,6trimethyl7(4nitrophenyl)2,4,6heptatrienylidene]2furanyl], 4Hpyran4one.
