摘 要:用双水相萃取(ATPE)丙酰螺旋霉素(PRO-SPM),分别用PRO-SPM溶液和发酵液,对聚乙二醇(PEG)和磷酸盐成相系统进行了研究,选出了合适的成相系统。在研究中发现PRO-SPM对成相有一定的影响。根据数据提出在PRO-SPM的双水相萃取过程中疏水作用是影响萃取的主要因素,与蛋白质以静电效应为主不同。用发酵液进行放大试验并与溶媒法进行比较,ATPE法萃取收率提高了32%,而纯度有所下降。初步探讨了聚合物的重复使用。The extraction technique of propionylspiramycin (PROSPM) by aqueous twophase (ATPE) from the solution or the fermentation broth was discussed. The effect of pH, PEG molecular weight and concentration, salt concentration were investigated in the PEG/phosphate system. The optimum condition was obtained. Results showed that the hydrophobic interaction was the dominant effect contrary to the partition protein by ATPE. The solute itself i.e. PROSPM engaged in the ATPE. Comparing with that of the organic solvent extraction, the recovery was 32% higher by ATPE, but the purity of the sample was somewhat lower. The feasibility of repeated use of PEG was also investigated.
