摘 要:目的:深入探讨放烧复合伤对造血微环境损伤的机制。方法:采用流式细胞仪观察了5.0Gyγ线放射损伤、15%Ⅲ度体表面积烧伤和放烧复合伤小鼠骨髓基质细胞表达血管细胞粘附分子(VCAM-1)、纤维连接素(Fn)、层粘素(Ln)和Ⅳ型胶原(ColⅣ)等细胞外基质成分的变化。结果:在本实验条件下,随着伤后时间的延长,骨髓基质细胞表达VCAM-1、Fn、Ln和ColⅣ的水平具有以单烧组>正常组>复合伤组>单放组的趋势;单烧组具有以伤后3、7、14d最高,单放组具有以伤后3、7d最低的时效特点。结论:骨髓微环境基质细胞粘附功能的损伤可能是放烧复合伤造血功能障碍发生的重要机制之一。Objective: To investigate the mechanism of hematopoietic inductive microenvironment after combined radiationburn injury. Methods: After the mice were inflicted with simple radiation injury, simple burn injury or combined radiationburn injury, the expression of cell adhesion molecules including vascular cell adhesion molecule1 (VCAM1), fibronectin (Fn), laminin (Ln) and collagen Ⅳ (Col Ⅳ) on bone marrow stromal cells cultured in vitro was detected with flow cytometry. Results: In the mice, the expression level of the adhesion molecules by the bone marrow stromal cells decreased in order from simple burn injury, control, combined radiationburn injury to simple radiation injury. Meanwhile, the level is the highest from the 3rd to 14th day in mice with simple burn injury while it is the lowest from the 3rd to 7th day in those with simple radiation injury postinjury. Conclusion: The damage of adhesion molecule expression by the bone marrow hematopoietic inductive microenvironment after combined radiationburn injury may play an important role in occurrence of hematopoietic disorder after the injury.
