摘 要:目的:探讨孕妇口服不同剂量米非司酮对血染色体稳定性的影响。方法:选择符合研究条件的中孕引产妇女90例,服药前抽取30例孕妇静脉血作为对照。根据服用剂量不同分为3组,每组30例,分别服用米非司酮150、200、300mg后48h水囊引产,于胎儿娩出后抽取孕妇静脉血,进行培养分化,观察姊妹染色体单体交换(SCE)频率,并应用细胞分裂阻滞法进行微核(MN)试验。结果:150mg、200mg组与对照组SCE频率比较差异无统计学意义(P>0.05);300mg组与对照组比较差异有统计学意义(F=3.433,P<0.001);各组MN率均比较差异无统计学意义(P>0.05)。结论:小剂量米非司酮(150、200mg)引产,不引起孕妇SCE频率增高,但加大剂量有增高SCE频率趋势。≤300mg剂量不会引起孕妇体内微核率的改变。Objective: To study the impaction of different dosage of mifepristone on the chromosome stabilities of pregnant women. Methods : 90 pregnant women with induction of labour in second trimester were registered in this study, and 30 blood samples were collected before medicine administration as the control. 90 cases were divided into 3 groups according to the different dosage of mifepristone ( 150mg, 200mg and 300mg, respectively), with 30 cases in each group. Induction of labor with water bag was performed at 48h after mifepristone administration. Venous blood of pregnant women was collected after fetal disengagement and then cultured and differentiated. The frequency of sister chromatid exchange (SCE) was observed and micronucleus (MN) assay was operated using cytokinesis blocked method. Results: Compared with the con- trol group, there were no statistical differences of SCE frequencies of the 150mg group and 200mg group ( P 〉 0.05 ), respectively, while the difference between the 300mg group and the control group was statistically significant ( F = 3.433, P 〈 0. 001 ). And the differences of MN rates of each group were not statistically significant (P 〉 0.05 ). Conclusion: Low dose of mifepristone ( 150rag and 200mg) used in induced labor wont cause the increase of SCE frequency in pregnant women, while a raising tendency of SCE frequency ap- pears with the increasing of mifepristone dosage. 300rag and lower doses of mifepristone won t cause the change of MN rate in pregnant women.
