瘦素、胰岛素对OLETF大鼠心肌细胞GluT-4含量的影响

摘  要:目的探讨瘦素、胰岛素与OLETF大鼠心肌胰岛素抵抗的关系。方法培养的12例大鼠心肌细胞分为11组,Western-blot方法检测不同浓度胰岛素、瘦素刺激不同时间葡萄糖转运子4(GLUT-4)转位效应的差异。结果瘦素和胰岛素促进GluT-4转位作用分别增加了2.28、3.14、3.07、9.06倍。胰岛素促GluT-4转位最佳时间为30min,瘦素最大效应浓度为100nmol/L,最佳刺激时间为30、60min。在OLETF大鼠,联合组GluT-4转位小于单纯胰岛素组,胰岛素组促进GluT-4转位作用也仅为对照组的69%。P均<0.05。结论胰岛素和瘦素均促进GluT-4转位,呈时间剂量依赖性;OLEFT大鼠心肌细胞存在胰岛素作用障碍,且瘦素与胰岛素联合应用可影响胰岛素的作用。Objective To study the relationship of insulin and leptin with GLUT-4 transloeation in OLETF rat cardiomyocyte. Methods The cardiomyocytes were incubated with leptin at various concentrations and times. The changes of GLUT-4 proteins were deteced by Western-blot. Results Leptin and insulin increased GLUT-4 translocation in OLETF rat eardiomyoeytes by 2.28 to 9.06 folds, respectively(P〈0.05). Meanwhile the most promotion of GLUT-4 transloeation were surveyed at 30rain after insulin inducement(P〈0. 001) and at 100nmol/1 and 30,60 min by leptin (P〈0. 05),respectively. In addition,the GLUT-4 translocation in OLETF rat cardiomyoeytes incubated in insulin plus leptin together were less than that treated with insulin alone (P 〈 0. 001). Moreover, the insulin-induced GLUT-4 translocation in OLETF rats was only 69% of the control rats (P〈0.05). Conclusions Both insulin and leptin can enhance GLUT-4 translocation in the manners of time and concentration dependences, and can restrain insulin-induced GLUT-4 translocation impairment.
