摘 要:目的观察水芹总酚酸(OJTPA)对α-萘异硫氰酸酯(ANIT)所致大鼠黄疸模型的退黄作用。方法将Wistar大鼠随机分为6组:正常组、ANIT模型对照组、茵栀黄颗粒阳性药物组、OJTPA高、中、低(300、150、75mg.kg-1)3个剂量组。各给药组连续给药7d,最后一次给药1h后,ANIT按75mg.kg-1体质量灌服大鼠诱发黄疸模型,正常组给等量的花生油。72h后取血取肝,检测血清总胆红素(T-Bil)含量以及肝组织中丙二醛(MDA)含量和超氧化物歧化酶(SOD)活力,同时观察肝脏组织病理学的改变。结果模型组大鼠血清T-Bil含量及肝脏MDA含量明显升高(P<0.01),肝脏SOD活性明显降低(P<0.01);OJT-PA能明显对抗这种改变,其退黄率分别为58.12%、43.12%、22.19%。肝脏组织病理学检查证实,OJTPA能减轻中毒大鼠肝细胞损伤程度。结论OJTPA退黄效果肯定,其机制可能与抗脂质过氧化有关。AImTo observe the jaundice-relieving effect of Total Phenolics Acid of Oenanthe Javanica (OJTPA) onα-Naphthylisothiocyanate (ANIT)-induced models of jaundice in rats. Methods Wistar rats were randomly divided into six groups : normal group , ANIT model group, Yinzhihuang control group and three OJTPA groups high ( 300mg·kg^-1) , middle ( 150mg·kg^-1 ) and low ( 75mg·kg^-1 ) dosage group. Intragastric administration of drugs were given to the Yinzhihuang control group and OJTPA groups for 1 week. At 1 hour after the last administration of OJTPA, five of groups were given ANIT (dissolved in peanut oil,75mg·kg^-1) by lavage and the normal group was given peanut oil alone by the same volume and in the same way. 72 hours after ANIT administration, the level of total bilirubin (T-Bil) in serum and malondialdehyde ( MDA ) content, superoxide dismutase (SOD) activity in hepatic tissue were detected, and histopathological changes of the liver were also observed. Results In the model group, the level of T-Bil in serum and MDA in hepatic tissue markedly increased after ANIT administration, and the level of liver tissue SOD also decreased significantly. OJTPA could obviously inhibit the alteration. OJTPA reduced the level of T-Bil significantly( the restrain rates of the high ,middle and low dosage groups were 58.12% 43.12% ,22.19% respectively). Pathological observation of the liver showed that OJTPA could relieve the degree of hepatic injury induced by ANIT in rats. Conclusion OJTPA can eliminate jaundice effectively. The mechanism is likely related to its anti-lipid peroxidation.
