摘  要:目的:建立同时测定椭圆叶花锚中4种主要酮类成分的HPLC方法。方法:采用Kromasil C18柱(4.6mm×300mm,5μm),流动相为甲醇-0.01mol.L-1磷酸氢二钾溶液(pH8.0)(80∶20),流速1.0mL.min-1,紫外检测波长269nm。结果:1-羟基-2,3,5-三甲氧基酮、1-羟基-2,3,4,5-四甲氧基酮、1-羟基-2,3,7-三甲氧基酮、1-羟基-2,3,4,7-四甲氧基酮进样量分别在0.0148~1.48μg(r=0.9999)、0.0296~2.96μg(r=0.9998)、0.0124~1.24μg(r=0.9995)、0.0136~1.36μg(r=0.9996)范围内线性关系良好。平均回收率(n=5)分别为101.0%,98.9%,97.9%,99.5%;RSD分别为2.2%,2.8%,1.4%,2.1%。结论:本法简便、快速、准确,重复性好,可作为椭圆叶花锚中的主要酮类成分的理想检测方法。Objective:To develop an HPLC method for simultaneous determination of four xanthones in Halenia ellipitica D.Don.Methods:The analysis was performed on Kromasil C18 (4.6 mm × 300 mm,5 μm) based on isocratic separation.The mobile phase consisted of methanol-0.01 mol·L^-1 dibasic potassium phosphate solution(pH 8.0) (80∶20) at a flow rate of 1.0 mL·min^-1 and the detection wavelength was 269 nm.Results:The linear ranges of 1-hydroxy-2,3,5-trimethoxyxanthone,and 1-hydroxy-2,3,4,5-tetramethoxyxanthone,1-hydroxy-2,3,7-trimethoxyxanthone,1-hydroxy-2,3,4,7-tetramethoxyxanthone were 0.0148^-1.48 μg (r=0.9999),0.0296-2.96 μg (r=0.9998),0.0124^-1.24 μg (r=0.9995),and 0.0136^-1.36 μg (r=0.9996) respectively;The average recoveries were 101.0%(RSD=2.2%),98.9%(RSD=2.8%),97.9%(RSD=1.4%) and 99.5%(RSD=2.1%),respectively.Conclusions:This method is simple,quick,sensitive,reliable and reproducible.It is the best choice for the determination of major xanthones in Halenia ellipitica D.Don.
