摘  要:目的:研究颅脑损伤后脑内Fos蛋白表达情况。方法:采用大鼠液压颅脑伤模型,应用免疫组织化学PAP法和计算机图像分析技术,观测不同程度颅脑损伤后不同时间脑内Fos蛋白的表达情况。结果:伤后15min脑内开始出现Fos蛋白表达,伤后1h达高峰,此后逐渐消退,伤后6h基本恢复至正常水平。Fos阳性神经元主要分布在损伤灶周围皮质、同侧梨状皮质和双侧海马齿状回,同侧海马较对侧显著。轻、中和重度脑损伤,Fos蛋白表达水平有显著差异,损伤越重,表达越强烈。结论:颅脑损伤引起脑内特定区域c-fos基因迅速和一过性表达增加,其表达水平与脑损伤程度有关。Objective: To study the induction of Fos protein in the brain of rats after traumatic brain injury. Methods: Using immunohistochemistry and computer image analysis, the induction of Fos protein in the brain of rats after fluid percussion brain injury in different degree was evaluated. Results: Robust Fos expression was observed in the ipsilateral cerebral cortex and bilateral hippocampus. The increased expression of Fos protein began 15 min after injury, peaked at 1 h postinjury, and subsided at 6 h postinjury. The intensity of label in the nuclei of Fos positive neurons and the numbers of Fos positive neurons were not equal after fluid percussion brain injury at different degree. The level of increased Fos protein following moderate brain injury was higher than that following mild injury (P<0.01), and the level following severe injury was higher than that following moderate injury (P<0.01). Conclusion: The pathobiologic changes at early intervals after traumatic brain injury may involve the alteration of cfos gene expression and cfos expression may be differentially regulated as a function of injury level.
