摘 要:目的:研究肿瘤坏死因子(TNF)对体外培养的牛脑微血管内皮细胞(BCMEC)与单核细胞(MNCs)粘附的影响及欧芹素乙(IMP)的拮抗作用。方法:通过测定MNCs中髓过氧化物酶活性的方法,对粘附于BCMEC单层上的MNCs进行定量。结果:TNF在5~250U/ml时呈剂量依赖性地诱导BCMEC与MNCs的粘附,TNF浓度为100U/ml时促粘附达最大值(61.0%)。TNF与内皮细胞共育1h,即可明显促进粘附作用(55.7%),2h时达最大刺激效应(62.8%)。IMP在浓度为0.1~10.0μmol/L时,呈剂量依赖性地拮抗TNF诱导的粘附作用,在10.0μmol/L时抑制率达90.3%。Objective: To study inhibitory effect of imperatorin(IMP) on tumor necrosis factor (TNF) induced adhesion reaction of cultured bovine cerebromicrovascular endothelial cell (BCMEC) with monocytes in vitro.Methods:The assay was based on the measurement of the myeloperoxidase activity in monocytes that had adhered to BCMEC layers.Results:At concentrations from 5 to 250 U/ml,TNF was shown to induce adhesion reaction of cultured BCMEC with monocytes in a dosedependent manner.The effect of TNF reached maximum (61.0%) at 100 U/ml.After 1 h incubation of BCMEC,addition of TNF significantly stimulated the adhesion reaction of BCMEC with monocytes(55.7%).After 2 h incubation the effect reached maximum(62.8%),then the effect slightly decreased.At concentrations from 0.1 to 10.0 μmol/L,IMP was found to antagonize induced adhesion reaction.Its maximal rate of inhibition was 90.3% at 10.0 μmol/L.Conclusion:IMP may decrease the TNF induced adhesion reaction of BCMEC with monocytes.
