摘  要:目的观察St.ThomasⅡ液对心肌微循环及内皮细胞(EC)的作用。方法制作离体心灌注模型,分缺血停跳组(A);单次灌注停跳液组(B);多次灌注停跳液组(C)。测定生理指标及毛细血管密度和充盈率;观察心肌及内皮细胞超微结构的改变。结果复跳后三组dp/dt均有下降,A组尤明显(P<0.01);五羟色胺只增加B、C组冠状血管流量(P<0.01);硝酸甘油增加冠状血管流量,B、C组多于A组(P<0.01)。停跳液可增加B、C组毛细血管充盈率(P<0.01)。结论内皮细胞依赖性舒张功能较非内皮依赖性舒张功能更易遭受缺血/再灌注损伤;St.ThomasⅡ液对内皮细胞有一定的保护作用,多次灌注优于单次灌注。Objective To investigate the protective effects of St. Thomas Ⅱ solution on the myocardial micro- circulation and endothelial cells. Methods Eighteen hearts obtained from SD rats were randomly and e- qually diveded into three groups and mounted on Langendorff circuits. Group A were subjected to 60 min ischemia, group B and C were protected by single cardioplegia infusion, multiple eardioplegia infusion, re- spectively, Endotnelium-dependent relaxation and endothelium-independent relaxation were recorded be-fore and after ischemia. The capillary competency and endothelium ultrastructure also were observed by light or electron microscopy. Results After ischemia, the dp/dt in group A was much decreased than in group B and group C (P<0.01); endothelium-dependent relaxations were damaged in group A. Compe-tency of capilearies in group B and C is more than that in group A (P<0.01). Both the myocardial and endothelial cell presented local pathological changes in group A. Slightly myocardial cells injuries and no signs of endothelial cell damage were found in group B and C. Conclusion St. thomas Ⅱ solution has somewhat benefit effects on endothelial cells against ischemia injury.
