人胚细胞的体外培养

摘　　要：目的：为医学研究长期提供培养的人胚细胞。方法：体外培养人胚细胞，观察其生长特性、染色体数目以及冻存和复苏的条件。结果：培养的人胚肺、皮肤、肾、脾及脑组织中，肺和皮肤细胞在冻存前后增长速度较快而稳定，染色体数目为２ｎ＝４６，占８７％～９１％；肾、脾细胞扩增缓慢；脑组织几乎不能扩增。结论：人胚肺和皮肤细胞可长期培养，可为医学研究提供二倍体细胞实验对象。Objective:To provide diploid cell strains of human embryonic tissues in vitro for medical study.Methods:Human embryonic tissue cells were cultured in vitro and their growth characteristics,cryopreservation,resuscitation and number of chromosome were studied.Results:Human embryonic tissues of lung,skin,kidney,spleen and brain were taken for cultivation.The growth of lung and skin cells was steady and fast before and after cryopreservation.The number of their chromosome in 2n=46 was 87%～91%.The multiplication of kidney and spleen cells was slowly and no growth appeared in brain cells.Conclusions:The diploid cell strains of human embryonic lung and skin have obtained which can undergo longterm cell divisions for medical study.
