摘 要:目的探讨血管生成与恶性胶质瘤生长之间的关系,为进一步抗血管生成治疗恶性胶质瘤提供实验依据。方法立体定向尾状核注射C6细胞制作SD大鼠脑胶质瘤动物模型并观察肿瘤生长变化;采用SABC免疫组化法,检测注射C6细胞后第9、15、21天肿瘤组织中增殖细胞核抗原(proliferating cell nuclear antigen,PCNA),检测CD34计算肿瘤组织中的微血管密度(microscope vessel density,MVD)。结果接种C6细胞后,随着天数的增加肿瘤体积逐渐增大;肿瘤组织中PCNA的表达逐渐增强,微血管密度明显增加,与对照组相比差异均有统计学意义(P<0.01,P<0.05)。结论恶性胶质瘤是生长活跃、血管增殖明显的肿瘤,血管生成与恶性胶质瘤生长间密切相关。Objective To approach the relationship between angiogenesis and growth of malignant glioma in rats, which was aimed to provide evidences for establish new anti - antigenic treatment strategies for the future. Methods A glioma model was established by injection of C6 cells into the right caudate nucleus of Sprague - Dawley rats stereotaxicly, and growth of tumor were observed. The proliferating cell nuclear antigen (PCNA) and microscope vessel density (MVD) were detected by immunohisto - chemistry at the 9th, 15th, 21st days after the C6 cells injected. Results The size of tumor increased gradually after C6 cells inoculated. PCNA and MYD were also increased after C6 cells inoculated, there were significant differences between control group and tumor groups (P 〈 0.01 ; P 〈 0.05 respectively). Conclusion Malignant glioma is a tumor of active growth and significant angiogenesis, and the angiogenesis is associated with growth of malignant glioma.
