摘 要:目的建立检测小鼠血清中刚地弓形虫NTPase-Ⅱ特异性IgGI、gM抗体的间接ELISA法,探讨用于弓形虫感染早期检测的应用价值。方法以刚地弓形虫NTPase-Ⅱ融合蛋白为包被抗原建立间接ELISA法,动态检测刚地弓形虫实验感染鼠血清NTPase-Ⅱ特异性IgGI、gM抗体变化。试验设脑囊虫病、血吸虫病及肺吸虫病病人血清对照。结果小鼠血清中的抗NTPase-Ⅱ抗体IgM于弓形虫感染后第3 d开始出现阳性反应,IgG抗体于感染后第7 d开始出现阳性反应,脑囊虫病、血吸虫病及肺吸虫病病人血清特异性IgG阳性率分别为0、1.92%和0。结论间接ELISA检测弓形虫感染鼠血清NTPase-Ⅱ抗体的敏感性和特异性较高,有望应用于人感染弓形虫的早期检测。Objective To detect IgG and IgM antibodies of Toxoplasma gondii NTPase-Ⅱby indirect ELISA and to evaluate the applied value of indirect ELISA thus established in the early infection of T.gondii.Methods Indirect ELISA in which T.gondii NTPase-Ⅱ fusion protein was used as an envelope antigen was established and was used to detect variation in NTPase-Ⅱ-specific IgG and IgM antibodies of T.gondii-infected mice.The sera of patients with brain cysticercosis,schistosomiasis,and paragonimiasis were used as a reference.Results IgM antibody was detected on the third day and IgG antibody was detected on the seventh day in mice blood serum infected with T.gondii.The positive rate of the IgG antibody in sera of patients with brain cysticercosis,schistosomiasis,and paragonimiasis was 0,1.92%,and 0.Conclusion The indirect ELISA thus established has satisfactory sensitivity and specificity and can be used to detect early infection of T.gondii.
