摘  要:目的观察人类MII期卵母细胞冻融过程中纺锤体及透明带的变化。方法 50例非男性因素的不育妇女,将常规体外受精后未受精的MII期卵母细胞进行慢速冷冻/快速复苏,使用Polscope系统观察冻融过程中纺锤体及透明带的变化;将复苏存活的卵母细胞分别培养1、3、5h后观察纺锤体的变化。结果 95枚未受精卵中52枚冷冻前和冷冻过程中均能观察到纺锤体。卵母细胞进入复苏-1液后47.8%可观察到纺锤体,后逐渐消失。46枚复苏存活卵母细胞分三组体外培养,1h纺锤体检出率为40.8%;3h为100.0%;5h为88.1%,差异具统计学意义。另外,冻融前后卵母细胞透明带的双折射性也发生了改变。结论卵母细胞的冻融过程会引起纺锤体及透明带的损伤,卵母细胞复苏后纺锤体进行重建。复苏后培养3h左右纺锤体的重建率最高。Objective.To observe the meiotic spindle and zona pellucida changes in living metaphase Ⅱ (MⅡ) human oocytes during the freezing and thawing procedures. Methods.. The unfertilized human oocytes were cryopreserved with a slow freezing/rapid thawing programme. Meiotic spindles and zona pellucida were analyzed at sequential steps of the freezing and thawing procedures with the use of Polscope system. And post-thawing living oocytes were observed at 1 h,3 h,5 h of culture respectively. Results:Meiotic spindles were observed in 52 MⅡ unfertilized human oocytes before and during the freezing procedure. Immediately after thawing of the frozen oocytes in solution-1, meiotic spindles were observed in 47.8% of oocytes, then gradually disappeared. For the post-thawing living oocytes, the rates of spindle observed were 40.8%, 100.0%, 88.1% at 1 h,3 h,5 h of culture, respectively. There were statistically significant differences of the spindle rates among the three groups. Meanwhile, zona pellucida changes were also observed during the freezing and thawing procedures. Conclusions:The current techniques for oocyte freezing and thawing inevitably caused the destruction of meiotic spindle and zona pellucida. The reconstruction of spindles was observed in all thawed oocytes. The spindle reconstruction reached the highest level at 3h after thawing.
